Cellular & Molecular Mechanisms of Cell Death - Cancers and Neurosciences

John Moores Liverpool University
(4/5) - 1 reviews

Liverpool John Moores University, Liverpool, UK

Main Areas of Interest

The department's research focuses on elucidating the cellular and molecular mechanisms underlying disease conditions characterised by either excessive cell death, e.g., neurodegenerative diseases (such as Parkinson's Disease and Alzheimer's Disease), or impaired cell death (such as cancers), with a view to harnessing the knowledge to develop novel therapeutics that will be more efficacious and elicit less side effects in managing those conditions. He uses state-of-the-art methodologies in pharmacology, physiology, biochemistry, cell and molecular biology, imaging, structural biology (through collaboration) and bioinformatics (through collaboration). He currently has collaborations with colleagues in the UK, Finland, South Africa and Nigeria.

His work is significantly translational (bench to bedside) and to some degree captures a wider context beyond cell death perturbations to assist in developing and optimising phenotypic- and target-based assays for high-throughput screening (HTS) of small molecules to identify those acting at a (druggable) molecular target of interest.

In summary, his current research interests include:

  • Exploring cellular and molecular mechanisms of cell death in neurodegeneration.
  • Oxidative stress (and nitrosative stress) and antioxidant signalling, including the Nrf2/ARE signalling pathway.
  • Prospecting for novel anti-cancer drug leads.
  • Development and optimisation of phenotypic and target-based assays for high-throughput screening (HTS) of chemical libraries (for various drug targets), HTS and compound characterisation/optimisation.
  • Development of novel screening platforms and technologies, including the use of 3D cell culture models, stem cells and patient-derived cells.
  • Natural products pharmacology and toxicology.
  • Drug repurposing/repositioning.
  • Use of fluorescent ligands to study novel pharmacology.

  • The use of cell culture in disease modelling and therapeutics development
  • Viability assays (absorbance, fluorescence or luminescence readouts), death assays for apoptosis, necrosis, parthanatos, etc.
  • Pharmacological assays: calcium assays, ligand-receptor interaction assays, reporter assays, etc.
  • Flow cytometry
  • Microscopy and imaging: brightfield and phase contrast microscopy, fluorescence imaging (wide-field, confocal)
  • Molecular biology techniques: transfections, gel electrophoresis, immunoblotting, PCR, siRNA technology, etc.
  • Immunocytochemistry
  • Enzyme-linked immunosorbent assays (ELISAs)
  • High-throughput screening of chemical libraries
  • GraphPad analysis of data

12 WEEKS PROGRAM

Fellows will be supported to settle down into their new environment (find appropriate accommodation, locate amenities, etc.) and register at the University, during which an ID card will be issued. A desk space will be allocated. Full induction (safety, laboratory practices, personal welfare, etc.) will be provided by the University’s designated staff and by myself. I will arrange an initial meeting with the fellow to discuss the objectives of the training and the expected mechanisms of their fulfilment. Experimental plans with realistic timelines will then be decided.

The fellow will be fully trained to run each experiment. They will then be allowed to attempt the experiment themselves, with the trainer present during their first run. They will be able to engage with me (supervisor) and other members of the laboratory to discuss any aspects of the general running of the laboratory or specific protocols that they require clarifications on. They will also be trained in how to analyse and interpret their data. I will arrange a weekly or bi-weekly meeting with the fellow to discuss and reflect on their progress and decide the next set of experiments based on data generated. A fellow must keep adequate and accurate records of their work using the laboratory book provided.

The fellow will be given opportunity to partake in research group meetings and other academic activities within the School and the University, including attending relevant bespoke workshops on research career development (e.g., on manuscript writing, grant writing, networking, etc.). They will also be encouraged to present their work orally at a research group meeting at least once before they complete their visit.

Towards the end of the training I will have a meeting with the fellow to discuss plans to publish their work (if enough data collected) or disseminate their findings in a future conference. They will also be assisted to identify existing or future opportunities, globally or within their original work environment, that the training has empowered them to be eligible and competitive for and how they can be assisted to access them and also integrate the knowledge and skills gained into their further research or professional practice roles.

All throughout the training any peculiar challenges or specific requests of a fellow will be fully considered to ensure a responsive, relevant and adequate training plan is developed for them.

This program allows Merit Applications. This program allows merit-based applications for virtual and onsite clinical and research programs. If you are successfully awarded under this category, Trialect or the host mentor will cover the tuition fee only. All applications will be evaluated based on merit. Due to the high level of competition, the chances of being selected under the merit category are quite limited.

Physicians and biomedical scientists are welcome to apply

The program allowed me to learn new techniques necessary for my PhD program and life-long research. It came at the right time just before starting my laboratory work. It also allowed me attend an international conference which was hosted by my host school, Liverpool John Moore and this helped in making connections with researchers in my field. My host, Dr. Amos Fatokun was more than just a host. He was like a father and ever ready to attend to even the minute questions. He was available and introduced me to other PhD and post doc students which made the training more bearable. Learning under him will remain one of the highest moments in my career as a researcher. However, fellows will benefit more if the period could be increased to minimum of 4 months. This is because first month is usually for traveling, settling into new environment and inductions and the remaining 3 months can be used for the training. Also, a full scholarship will enable more researchers to benefit from these traineeships. A lot of young researchers do not have sponsors, so when given discounted merit, we have to look for sponsors and most times we lose the opportunity because of inability to get sponsors. For instance, I would not have benefited from this Trialect program without my sponsors; Consortium for Advanced Research raining in Africa (CARTA), therefore, I am forever grateful. How to do better in the next traineeship: I will say by giving 100% scholarship.
