The research interests in my laboratory are focussed on the growth and signaling factors, and particularly those secreted by macrophages, in the muscle regeneration processes. In particular, we are studying how the factors produced in the different phases of the inflammatory response that accompanies muscle repair affect the proliferation and differentiation capabilities of the bona fide myogenic cells (satellite cells), of the fibro-adipogenic progenitors and of the pericytes. These latter have recently been shown to possess good myogenic potential and although they are not part of the physiological regeneration process they represent a promising candidate in muscle-related regenerative medicine. Other collaborative projects presently carried out in the laboratory include: studying the effect of the macrophagic factors in age-related muscle atrophy, the creation of 3D implantable muscle constructs to treat congenital malformations, the repurposing of anti-oxidant drugs in the context of dystrophic muscle to improve the mitochondrial loss of functionality present in DMD patients.

Isolation of stem cells from murine muscles (satellite cells, fibro-adipogenic progenitors, pericytes) using Miltenyi magnetic beads, from adult and senescent animals
In vitro culturing and differentiation of above-mentioned cells.
Harvesting and processing of specific muscles for the preparation and culturing of viable single muscle fibers
Assays of cell viability, proliferation and apoptosis.
Measurements of intracellular ROS levels and mitochondrial potential by means of specific fluorescent probes.
Immunofluorescence assays of cultured cells and tissue sections, both with epifluorescence and confocal microscopy.
Expression analysis of specific myogenic markers by RT-PCR and western blot.
Culturing of murine macrophagic cell lines and their use to prepare conditioned medium.
