Main Areas of Interest
Research in my laboratory focuses on the pathogenic mechanisms of cancer with particular focus on liver cancer. We are investigating the mechanisms underlying processes that sustain tumor progression such as tumor-stromal interactions, tumor infiltration and metastatic dissemination. The goal is to elucidate mechanisms driving cancer progression, with the ultimate goal of identifying targets for developing drugs that interfere with the evolution of the malignant process. Major research interests include primary and secondary hepatic tumors: i) pathogenesis of hepatocellular carcinoma (i.e. tumor-stromal interactions, invasion and metastasis); ii) the role of the tumor microenvironment in the development and growth of liver metastasis; iii) identification of molecular therapeutic targets and development of new drugs in liver cancer.
The main investigated molecular determinants include tetraspanins, integrine & ECM proteins and proteases. More recently, my research interest focuses on molecular mechanisms involving lysophosphatidic acid (LPA) in mediating both the paracrine interactions between stromal fibroblasts and cancer cells and the autocrine self-sustaining tumor growth supported by LPA receptor 6 (LPAR6), which we have recently characterized. The LPA signaling pathway is an attractive target for pharmacological interventions in HCC on which we are currently working.

- 2D and 3D cell culture using: human and murine chemo-immunosensitive and chemo-immunoresistant tumor cells; primary tumor samples (Hepatocarcinoma cell lines; Colony formation assays
- Cell culture and transformation (gene silencing, gene: overexpression)
- Cell co-culture models: HCC cells and hepatic stromal cells such as carcinoma-associate fibroblasts and hepatic stellate cells)
- Cell viability/toxicity assays (necrosis, apoptosis and senescence, cell cycle analysis)
- Cell metabolism assays (enzymatic activity by spectrofluorimetric and chemiluminescencse-based assays; metabolic radiolabeling)
- Chromatography (thin layer chromatography, ion exchange chromatography, affinity chromatography)
- Immunoblotting, co-immunoprecipitation
- Cell sub-fractioning, plasma-membrane isolation proteins (mitochondria isolation and related functional activities)
- Qualitative and quantitative PCR; PCR-arrays
- Chromatin immunoprecipitation (ChIP)
- ELISA
- Flow cytometry
- Fluorescence and confocal microscope
- Immunohistochemistry
- Immunological assays: tumor cell phagocytosis, immune-phenotyping of immune cells and tumor cells (flow cytometry/ELISA)
- Oncological mouse models: immunocompetent and immunodeficient mouse models of HCC; mice handling, tumor monitoring, drug administration, ex vivo immune system activity of murine models.
Laboratory techniques include cell and tissue culture, cellular & molecular biology techniques, biochemical and pharmacological techniques. Correlated research interests include: i) contribution of liver cirrhosis to cancer formation; ii) heterogeneity in liver cancer; iii) alteration of microenvironment in liver cancer; iv) identification of molecular therapeutic targets and development of new drugs.
