Research lines

The main research areas of our group are:

  • molecular basis of onset and reversion of chemoresistance in human tumors;
  • molecular basis of onset and reversion of immune-resistance/immune-evasion in human tumors
  • synthesis and validation of new combinatorial antitumor therapies (e.g. multitarget drugs; nanoparticles-based chemotherapy; chemo-immunotherapy approaches).

 

Experimental techniques

  • 2D and 3D cell culture using: human and murine chemo-immunosensitive and chemo-immunoresistant tumor cells; primary tumor samples (mesothelioma, glioblastoma, leukemias); primary culture of tumor stem cells
  • cell culture and transformation (gene silencing, gene: overexpression)
  • cell co-culture models: brain-blood barrier/glioblastoma cells; peripheral blood mononuclear cells/tumor cells
  • cell viability/toxicity assays (necrosis, apoptosis, cell cycle analysis, DNA genotoxicity, autophagy, senescence)
  • cell metabolism assays (enzymatic activity by spectrofluorimetric and chemiluminescencse-based assays; metabolic radiolabeling)
  • chromatography (thin layer chromatography, ion exchange chromatography, affinity chromatography)
  • immunoblotting, co-immunoprecipitation
  • cell subfractioning, plasma-membrane isolation proteins
  • qualitative and quantitative PCR; PCR-arrays
  • chromatin immunoprecipitation (ChIP)
  • ELISA
  • flow cytometry
  • fluorescence and confocal microscope
  • immunohistochemistry
  • immunological assays: tumor cell phagocytosis, anti-tumor lymphocyte T expansion and activity, immune-phenotyping of immune cells and tumor cells (flow cytometry/ELISA)
  • oncological mouse models: immunocompetent mouse models of breast cancer, lung cancer, osteosarcoma, pleural mesothelioma; mice handling, tumor monitoring, drug administration, ex vivo immune system activity of murine models.

This program allows Merit Applications. This program allows merit-based applications for virtual and onsite clinical and research programs. It was a great experience with Prof. Chiara Rignati, Torino University. She personally took interest to make sure I gained from the 2 weeks' experience at her lab. The facilities are outstanding, and the ways were friendly. She was easily accessible throughout the training perdiod, despite her busy schedule. I only wish I could have spent more time, so that I could benefit more.
