Trialect at https://trialect.com/, a US incorporation, has partnered with the faculty at the University of Torino, Italy, to provide short term hands-on traineeship/fellowship (6-12 weeks) in the area of enzyme biochemistry with specific emphasis on in vitro drug metabolism. The lab offering you the traineeship is one of the leading labs in this field and will help you co-author papers in the areas of Protein Engineering, Drug Metabolism, Biocatalysis, Biosensing and Nanotechnology. Only 3 positions are available. We appreciate if you can refer your students/colleagues/fellows (physicians/biomedical scientists) who may be interested in the program. 

The Enzyme Biochemistry Experimental techniques you will have the opportunity to learn during your program include the following:

  • Bacterial transformation, clone design (gene: cloning, mutagenesis, evolution);
  • Bacteria cell culture using: E. coli expression systems (2L flask, 5L fermentation);
  • Whole-cell assays: use of bacterial whole cells for screening drug candidates or the production of synthetic intermediates (cell culture, multi titer plate reader, HPLC)
  • Protein isolation: (cell lysis with sonication, ultracentrifugation, chromatography)
  • Protein purification using chromatography (ion exchange, affinity and size-exclusion)
  • Enzyme catalysis (spectrofluorimetric, chemiluminescence-based assays; HPLC separation and identification assays)
  • Enzyme kinetics (stopped-flow spectroscopy, fluorescent-based assays)
  • Protein stability (FT-IR, Differential Scanning Calorimetry, unfolding studies using fluorescence and Circular Dichroism)
  • Protein-ligand interactions (Differential Scanning Calorimetry, Isothermal Titration Calorimetry, Fluorescence)
  • Qualitative PCR for cloning/subcloning purposes
  • Protein immobilization for bioelectrochemical platforms (carbon/gold/gold nanoparticles and graphene)
  • ELISA
  • X-Ray crystallography (screening and optimization of protein crystal conditions using dedicated kits)
  • In silico Molecular modelling and Docking (homology model construction, optimization, validation, enzyme-substrate/inhibitor docking)
  • Drug metabolism studies: in vitro assays for approved drugs or new chemical entities, drug-drug and drug-food interactions, screening, and production of single nucleotide polymorphic protein variants; protein characterization: enzyme activity, structure-function relationship, stability, and immobilization.

This program allows Merit Applications. This program allows merit-based applications for virtual and onsite clinical and research programs. If you are successfully awarded under this category, Trialect or the host mentor will cover the tuition fee only. All applications will be evaluated based on merit. Due to the high level of competition, the chances of being selected under the merit category are quite limited.
